A new method for constructing three-dimensional ratio type fluorescent probes by Zhu Liangliang

[ China Instrument Network Instrument Development ] Fluorescent signal visualization sensing is one of the important methods in chemical and physical detection. In addition to identifying target substances in traditional applications, the application of fluorescent probes to the microenvironment to detect environmental behavior changes in chemical or life science processes is also highly desirable. Recently, Zhu Liangliang, a researcher in the Department of Polymer Science at Fudan University, used a single-molecule fluorescence-delayed fluorescent double emission to construct a new method for three-dimensional ratio-type fluorescent probes. Related research results were published on February 13th in Nature Communications. The researchers have further developed a 3-D ratio luminescence sensing system for the precise detection of polarity changes in complex phospholipid systems, which is expected to provide new ideas for the diagnosis of membranous lesions.

According to reports, most fluorescent probes work by responding to changes in a single transmitted signal. In practical applications, factors such as changes in probe concentration or fluid properties can easily cause the reading of an erroneous signal. In recent years, significant progress has been made in solving these interference factors by fluorescence correction methods, such as self-calibration using the characteristics of dual emission fluorescence. However, the use of only fluorescent emissions is easily overlapped with the signal of the self-fluorescent material of many environmental media. Therefore, simultaneously overcoming these interference factors has become a big problem in exploring and developing new fluorescence technologies.
Recently, Zhu Liangliang's group proposed a new method to achieve fluorescence (FL)-thermal activation delayed fluorescence (TADF) dual emission behavior on a single molecule to create a new probe analysis technology. TADF promotes the development of organic electronics as a metal-free induced long-lived emission, known as the third-generation OLED material. Zhu Liangliang said, "We hope to design a new type of sensor using the molecular dual emission strategy of the simultaneous change of the emission wavelength and lifetime of the TADF signal and the relative constancy of FL." Taking the polarity change of the induced environment as an example, TADF is used as the sensing signal and its emission wavelength. And the lifetime changes with the change of the environment polarity; FL as the internal standard reference signal, its wavelength and lifetime do not change with the environmental polarity. Zhu Liangliang's group further established a three-dimensional ratio luminescence sensing system: environmental polarity (X-axis), ratio wavelength (Y-axis), and ratio lifetime (Z-axis). Compared with the traditional two-dimensional curve application, the construction of the three-dimensional analysis system greatly reduces the measurement error and improves the application accuracy.
Li Xuping, a doctoral student in the Department of Polymer Science at Fudan University, is the first author of the paper. Theoretical calculations and spectral analysis were partially assisted by the Royal Institute of Technology (KTH) and the Polymer Science Department of Zhejiang University. The related work also applied for a patent for invention (CN2018100470946).
